it was the episode Route 666 in Season One. Her name was Cassandra and her and Dean used to go out. Dean told her what he did for a living and she called him with a case for him and Sammy to investigate

The character's name was Cassandra...she was the one non-hunter person that Dean actually told what he did for a living.
