Given two points (x1,y1) and (x2,y2), the equation of the line through the two points is

(y - y1) / (x - x1) = (y2 - y1) / (x2 - x1)

Generally speaking, the line of best fit is the line which minimizes the mean square error between the line and the set of points provided. Since two points determine a line uniquely, the mean square error between the equation of the line (above) and the two given points is necessarily zero. So the equation of the line is also the equation of the line of best fit.
