You don't state what you wish to do with A3:S3 if BOTH A3 and B3 have an 'x'.  So, these rules will simply not format any fill color if that condition exists.

    Highlight A3:S3

In the Home tab, select Conditional Formatting and click 'New Rule'

Select 'Use a formula to determine which cells to format'.

In the 'Format values where this formula is true' textbox, enter this formula:

    =AND($B$3="x",$A$3<>"x")

Click 'Format', select the Fill tab, select the Red option, and click 'OK' twice.

Repeat the above process, except using this formula and selecting the Yellow option:

    =AND($A$3="x",$B$3<>"x")

you need to set 2 conditionals for that. one for red, one for yellow.

go to the home tab and use conditional formatting, set up the rules to do what you need
